2-ddCt Method
The Livak method computes ddCt = (Cq_target - Cq_reference)_sample - (Cq_target - Cq_reference)_calibrator and reports fold change as 2^-ddCt. The base of 2 hard-codes the assumption that target and reference both amplify at exactly 100% efficiency, so an untested efficiency difference of a few percent compounds across cycles into a large error in reported fold change.
