Peptide bonds reduce Cu2+ to Cu+ in alkaline conditions, and bicinchoninic acid chelates Cu+ to give a purple complex read at 562 nm against a BSA standard curve. It tolerates detergents such as SDS and Triton, unlike Bradford, but is inhibited by reducing agents like DTT and by chelators such as EDTA.
