Nalpha-Boc is removed by TFA each cycle and benzyl-type side-chain groups plus the linker survive until final cleavage with anhydrous HF or TFMSA, so the strategy is graded-acid rather than orthogonal. It remains the method of choice for aggregation-prone and thioester-containing peptides because repeated TFA treatment disrupts on-resin hydrogen bonding.
