Cas9 Nickase

Mutating one of Cas9's two nuclease domains (D10A or H840A) leaves an enzyme that cuts only a single strand. The nick is the foundation of base and prime editing: it directs the cell's repair machinery toward the edited strand without creating a double-strand break.
