Chromogenic Detection with DAB

HRP oxidizes diaminobenzidine in the presence of hydrogen peroxide into an insoluble brown polymer at the antigen site; AEC (red) and alkaline-phosphatase chromogens such as Fast Red serve as alternatives when tissue contains melanin or hemosiderin. DAB development time is a nonlinear amplification step and must be timed identically across a run.
