Shaking the sample with D2O replaces exchangeable OH, NH, and COOH protons with deuterium, making their 1H signals vanish on re-acquisition. These protons also show concentration- and temperature-dependent shifts and often broad or absent coupling because exchange is fast on the NMR timescale.
