Whole-slide images are color-deconvolved into hematoxylin and DAB channels, cells are segmented (increasingly by trained neural networks), and per-cell intensity and positivity are computed to produce a continuous, operator-independent score. Stain normalization across scanners and batches is required before scores from different runs can be compared.
