HRP oxidizes luminol in the presence of peroxide and an enhancer, emitting 425 nm light captured by a CCD imager or film. Signal is enzymatic and therefore time-dependent and non-linear: it peaks and decays over minutes, which gives high sensitivity but a narrow, substrate-dependent quantitative range.
