Formalin Fixation Cross-Linking Chemistry

Formaldehyde forms reactive methylol groups on lysine, arginine and other nucleophilic side chains, which condense into methylene bridges that covalently tie neighboring proteins together. The reaction is slow, time- and temperature-dependent, and partially reversible by heat, which is why fixation duration governs downstream antigenicity.
