Direct labeling puts the reporter on the primary antibody; indirect adds a labeled secondary for amplification; ABC exploits avidin-biotin-peroxidase complexes; and modern polymer systems attach many HRP molecules to a dextran or micropolymer backbone for high sensitivity with low background and no biotin. Sensitivity gains trade against increased risk of amplifying nonspecific binding.
