A valid run includes known positive tissue, known negative tissue, an omitted-primary control that exposes detection-system background, and an isotype or concentration-matched irrelevant-antibody control that exposes nonspecific immunoglobulin binding. Internal positive cells within the test section are the strongest evidence the run worked on that specific slide.
