After amplification with an intercalating dye, the instrument ramps temperature and records the negative derivative of fluorescence, producing a peak at each product's melting temperature. A single sharp peak indicates one specific product; extra peaks or a low-temperature shoulder reveal primer-dimers or non-specific amplicons that the dye was counting as signal.
