Membrane Blocking

Unoccupied membrane surface adsorbs antibody nonspecifically, so it is saturated with an inert protein or polymer before probing. Nonfat milk is cheap but contains casein, a phosphoprotein that raises background and can sequester phospho-specific antibodies, so BSA or a defined synthetic blocker is used for phospho-targets and for biotin/streptavidin systems.
