A peptide C-terminal thioester and a second peptide with an N-terminal cysteine undergo reversible transthioesterification followed by an irreversible intramolecular S-to-N acyl shift, forming a native amide bond chemoselectively in aqueous buffer with unprotected side chains. Desulfurization or thiolated auxiliaries extend it to non-Cys junctions, putting 100+ residue proteins within reach.
