Prime Editing

A Cas9 nickase fused to a reverse transcriptase, guided by a pegRNA that both targets the site and encodes the desired edit, writes new sequence directly into the nicked strand. It installs all twelve base substitutions plus small insertions and deletions without a double-strand break or donor DNA.
