Protospacer Adjacent Motif

Cas9 will not cut unless a short PAM sequence (5'-NGG-3' for SpCas9) sits immediately 3' of the protospacer on the non-target strand. The PAM licenses DNA unwinding, is what keeps bacteria from cutting their own CRISPR array, and is the hard constraint on which genomic positions are targetable.
