Reverse Transcription

Reverse transcriptase uses an RNA template plus a primer (oligo-dT, random hexamer or gene-specific) to synthesize a complementary DNA strand, with RNase H activity degrading the RNA in the hybrid. Every short-read RNA-seq protocol sequences cDNA, not RNA.
