Signal Saturation and Dynamic Range
Once a pixel reaches the detector's maximum value the band's true intensity is lost, so a saturated band under-reports abundance and flattens real differences. Correct practice is to collect a short exposure series, confirm no saturated pixels in the bands being compared, and verify that a dilution series of the lysate gives a linear intensity response.
