Full-spectrum cytometers record each event across dozens of detectors and solve a least-squares fit of the observed spectrum against reference signatures for every fluorophore. Because autofluorescence can be entered as its own signature and extracted, unmixing resolves dyes with near-identical peaks that conventional compensation cannot separate.
