Uronium Coupling Reagents
HBTU, HCTU and HATU convert the carboxylate into a benzotriazolyl or azabenzotriazolyl active ester in the presence of a tertiary amine base such as DIPEA; HATU is faster and lower-epimerizing because the pyridine nitrogen of HOAt provides neighbouring-group assistance. Uronium reagents must not be used in excess over the acid or they cap the resin amine as a guanidinium.
