A drop of protein mixed with reservoir solution is equilibrated against a larger reservoir; water leaves the drop as vapour until the two match in chemical potential, slowly concentrating both protein and precipitant. Hanging-drop and sitting-drop geometries trace a defined path across the phase diagram set by the initial drop-to-reservoir dilution.
