About Peptide Synthesis: From Resin Bead to Verified Sequence

You will learn how a peptide is built one residue at a time on an insoluble support, why Fmoc/tBu displaced Boc/Bzl as the default strategy, and how resin, linker, coupling reagent and protecting-group set are chosen together as an orthogonal system. You will be able to diagnose failed couplings, aggregation and epimerization, design a cleavage cocktail with the right scavengers for a given side-chain set, and purify and confirm the product by RP-HPLC and mass spectrometry. You will finish able to plan a route to peptides longer than stepwise SPPS can reach using native chemical ligation.
